Laccase Production from Local Biomass Using Solid State Fermentation

The large family of enzymes, known as polyphenols oxidases, includes laccase. Due to the inclusion a copper atom in their catalytic core, laccases are frequently referred multi-copper oxidases. Laccases versatile enzymes that can catalyze oxidation wide range phenolic and non-phenolic substances. In current study, local strain Aspergillus niger was used for solid-state fermentation produce fungal laccase, well purify optimize enzyme profile, which acquired using guaiacol measure activity, showed after five days incubation, wheat straw provided highest level laccase or 2.551 U/mL. A technique called response surface methodology (RSM) examine effects various conditions on production enzymes. RSM results demonstrated activity at its 45 °C, pH 5.5, 30% moisture level, inoculated with 2 mL mycelium. Through ammonium sulphate precipitation dialysis, purified. Additionally, column chromatography further next step characterization evaluate how temperature affected activity. At °C isolated produced findings study A. is capable producing an economical environmentally friendly way. unique oxidative features, this has received lot attention recently.
